The New York Panorama feature on Page 1 misidentifies the location of a picture that shows

The New York Panorama feature on Page 1 misidentifies the location of a picture that shows a tour guide photographing members of her group with the Statue of Liberty. It was Liberty Island, not Ellis Island. The On the Market article last Sunday described incorrectly the features of a house in Ho-Ho-Kus, N.J. There is an attic in the house. The cover story on April 7, about the development of Hudson Yards, described incorrectly the type of bonds sold to help begin development of Battery Park City. They were state-authorized bonds secured by a nonbinding agreement, not state-guaranteed bonds, which would have been secured by a legal obligation and less risky. An article on Page 64 about the acquisition of Remington by Cerberus misstates the birthplace of Stephen Feinberg, a founder of Cerberus. He was born in the Bronx, not Spring Valley, N.Y., which is where he grew up. An article on Page 54 about Meow Wolf, an art collective that creates immersive and interactive experiences, refers imprecisely to Creative Startups. It is a start-up accelerator, not a business incubator. An article on April 21 about gender-discrimination lawsuits against the Salk Institute misstated the locati...

Functional diversification of hybridoma-produced antibodies by CRISPR/HDR genomic engineering

↵* Present address: Department of Experimental Immunology, Amsterdam University Medical Center, Amsterdam, Netherlands. Hybridoma technology is instrumental for the development of novel antibody therapeutics and diagnostics. Recent preclinical and clinical studies highlight the importance of antibody isotype for therapeutic efficacy. However, since the sequence encoding the constant domains is fixed, tuning antibody function in hybridomas has been restricted. Here, we demonstrate a versatile CRISPR/HDR platform to rapidly engineer the constant immunoglobulin domains to obtain recombinant hybridomas, which secrete antibodies in the preferred format, species, and isotype. Using this platform, we obtained recombinant hybridomas secreting Fab′ fragments, isotype-switched chimeric antibodies, and Fc-silent mutants. These antibody products are stable, retain their antigen specificity, and display their intrinsic Fc-effector functions in vitro and in vivo. Furthermore, we can site-specifically attach cargo to these antibody products via chemoenzymatic modification. We believe that this versatile platform facilitates antibody engineering for the entire scientific community, empowering p...

Stax SRM-D50 Electrostatic Earspeaker Driver & DAC Review | – StereoNET Australia & New Zealand

In all my years as a headphone listening enthusiast, one thing has remained clear from the very start – there is nothing quite like listening to a Stax system. We take a closer look at the new Stax SRM-D50 which blends electrostatic earspeaker driving tradition with a modern DAC in one unit. There is something truly intoxicating behind every listen to a Stax earspeaker. It’s not just the sheer audio quality that is so captivating, but there is a certain charm shared by no other manufacturers that really make Stax products unique and special. To me, the headphone market is not dissimilar to the car market. There are high-end German performance pieces which look incredible, are beautifully packaged and boast top-tier performance. Then on the other end of the spectrum, there are budget-friendly releases, that might perform effectively for the price range, but fail to live up to long term expectations or suffer build quality issues. In this analogy, Stax is the completely stripped out race track car, which has shed all creature comforts such as air conditioning, a sound system, or passenger seats. Stax is all about sheer performance above everything else. And as a result...

They adorn the ends of Cat5 network patch cables and the flat satin cables that come

They adorn the ends of Cat5 network patch cables and the flat satin cables that come with all-in-one printers that we generally either toss in the scrap bin or throw away altogether. The blocky rectangular plugs, molded of clear plastic and holding gold-plated contacts, are known broadly as modular connectors. They and their socket counterparts have become ubiquitous components of the connected world over the last half-century or so, and unsurprisingly they had their start where so many other innovations began: from the need to manage the growth of the telephone network and reduce costs. Here’s how the modular connector got that way. For the first 80 years or so of the US telephone network, the Bell Company called all the shots. They owned absolutely every bit of equipment in the system – the wires on the poles, the switchgear in the central offices, microwave links, and even the phone sets in customer homes. They had complete control over every aspect of the delivery of their services, and used their monopoly to build an incredibly integrated and durable system. But with great power comes great responsibility, or in the case of a complex technological system prone to breakdown ...

Heating an entire home with electric heat is an expensive option, but for anyone who complains

Heating an entire home with electric heat is an expensive option, but for anyone who complains about routinely shivering in a certain room, or even a certain chair, the gift of an electric space heater might be warmly received. (Or buy one for yourself.) Here are a few top picks from Consumer Reports’ latest space heater tests. The latest crop of space heaters we’ve tested have improved safety features, such as a switch that shuts the heater off if it’s tipped over or it overheats. Space heaters, after all, still account for roughly one-third of all home heating fires and more than 80 percent of associated deaths, so you can’t buy one without doing a little homework.  And although many we tested were less hot to the touch than in the past and have plastic rather than metal grates, burns to the hands—particularly among kids—remain common. Best Electric Heater Manufacturers Cheap Price Vornado AVH2, $100 Fairly quiet running, fast heating, and a five-year warranty are among chief strengths of this electric heater. You also get mechanical controls mounted on top for easy accessibility, a plus since there’s no remote control, and multiple fan speeds. This unit also remained re...
